Product Specific Information

This 3A6 monoclonal antibody reacts with human CD166 (activated leukocyte cell adhesion molecule, ALCAM). CD166 is an immunoglobulin superfamily cell adhesion molecule expressed on hematopoietic (activated T/B cells, monocytes, dendritic cells) and non-hematopoietic tissues (neural, endothelial, epithelial).
It is detected in melanoma, colorectal carcinoma, breast, prostate, and bladder cancers. CD166 mediates cell-cell adhesion via ALCAM-ALCAM or ALCAM-CD6 interactions and plays roles in development, inflammation, and neutrophil migration. Its expression serves as a prognostic and cancer stem cell marker.

The 3A6 antibody induces internalization and recycling of CD166.

• Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.25 µg) per test.
  A test is defined as the antibody amount staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Super Bright 600 is a tandem dye excited at 405 nm and emits at 600 nm. Recommended filter: 610/20 bandpass.
Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright conjugates to reduce non-specific polymer interactions.
Light sensitivity: Protect vial and stained samples from light.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.
Excitation: 405 nm; Emission: 600 nm; Laser: Violet Laser.

Target Information

The ALCAM (CD166) gene encodes an immunoglobulin receptor with five Ig-like domains (VVC2C2C2). It binds to T-cell differentiation antigen CD6 and is involved in cell adhesion and migration. Multiple alternatively spliced transcript variants exist.
