Product Specific Information

This HMN3-133 monoclonal antibody reacts with mouse Notch3, one of four members of the Notch family of receptors. Notch receptors are 300-kDa single-pass transmembrane proteins. The extracellular domain contains epidermal growth factor-like repeats for ligand binding, while the intracellular domain participates in cell signaling.

Upon binding its membrane-bound ligand (Delta or Jagged), the Notch receptor undergoes proteolytic cleavage by ADAM-family metalloproteases and gamma-secretase. The released Notch intracellular domain (NICD) translocates into the nucleus, associates with RBP-J, recruits co-activators, and activates transcription.

Notch3 expression occurs in regulatory T cells, thymocyte subsets (CD4-CD8-, CD8SP), vascular smooth muscle, and the central nervous system. It plays a role in hematopoiesis, T cell lineage commitment, and thymocyte development, particularly in the DN3 (CD4-CD8-CD25+CD44-) stage. Notch3 may regulate the pre-TCR checkpoint via NFkappaB and E2A signaling. Overexpression of Notch3 has been implicated in leukemia.

A test is defined as the amount of antibody that stains cells in 100 µL volume, typically 10⁵–10⁸ cells/test.
Careful titration is recommended for optimal performance.

Target Information

NOTCH3 is a human homologue of the Drosophila Notch type I membrane protein. In Drosophila, Notch-ligand interactions (delta, serrate) regulate neural development. Human homologues of these ligands have been identified. NOTCH3 functions as a receptor for Jagged1, Jagged2, and Delta1, regulating cell-fate determination. Mutations in NOTCH3 are associated with CADASIL (Cerebral Autosomal Dominant Arteriopathy with Subcortical Infarcts and Leukoencephalopathy).
