Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly during late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus. Ki-67 shows cell cycle–dependent distribution: perinucleolar expression at G1, nuclear matrix localization at G2, and chromosomal association during M phase.
As a proliferation marker, Ki-67 is absent in G0 cells but increases steadily from G1 through mitosis. Ki-67 antibodies are valuable for assessing proliferative fractions in neoplasms, with prognostic relevance comparable to thymidine-labeling indices. A low Ki-67 index correlates with histologically low-grade tumors. It is also used as a neuronal marker of cell cycling and proliferation.
