Product Specific Information
This antibody is predicted to react with bovine, canine, amphibian, mouse, porcine, and rat based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 10 minutes at room temperature.
A suggested positive control is tonsil or ovarian carcinoma.
Target Information
Claudin-4 belongs to the 20-member claudin family, which consists of transmembrane cellular adhesion proteins localized at epithelial and endothelial tight junctions.
Claudins exhibit tissue-specific expression patterns and are essential for tight junction charge-specific channels regulating paracellular ion flux.
Claudin-4 contributes to tight junction strands and may play a role in internal organ development and function during pre- and postnatal life.
Overexpression of Claudin-4 decreases paracellular electrical conductance due to reduced Na⁺ permeability, without affecting Cl⁻ permeability.
In ovarian carcinoma cells, Claudin-3 and Claudin-4 are upregulated at both mRNA and protein levels. Claudin-4 is also overexpressed in pancreatic cancer tissues and other epithelial cancers.
The Claudin-4 gene is deleted in Williams-Beuren syndrome, a neurodevelopmental disorder affecting multiple systems.
