Target Information
TAK1 (MAP3K7)는 serine/threonine kinase 계열에 속하는 다기능 신호 단백질로, TGF-β 활성화 kinase로 처음 발견되었습니다. TAK1은 TGF-β receptor 및 TRAF6과 상호작용하여 JNK 및 p38을 조절하며, Wnt, BMP, activin, thyroid hormone, osmotic stress, endothelin, ephrine 등의 다양한 신호 경로에서도 활성화 역할을 합니다.
또한 TAK1은 von Hippel-Lindau tumor suppressor를 활성화하여 PDGF-B, integrin β1, β5를 억제함으로써 상처 치유를 촉진합니다. IL-1, IL-6, IL-18, TNF, CD40, TLR, RIG-I 등 다양한 경로에서 IKK 활성화에도 필수적이며, TAB2/3와의 결합 및 polyubiquitination을 통해 TAK1-IKK 경로가 활성화됩니다. TAK1은 Thr178, Thr184, Thr187, Ser192에서 자가 인산화되며, IKK-β를 인산화합니다. 또한 인간 telomerase reverse transcriptase를 억제하여 세포 수명 조절에 관여합니다.
