Target Information
HIF-1 beta는 aryl hydrocarbon receptor nuclear translocator (ARNT) 유전자의 산물로, hypoxia 관련 질환의 병태생리에 중요한 역할을 합니다. Hypoxia는 심근 및 뇌 허혈, 암, 폐고혈압, 선천성 심장질환, 만성 폐쇄성 폐질환 등 다양한 질환에 기여합니다.
HIF-1은 포유류의 산소 항상성 유지에 관여하는 핵 단백질로, prolyl hydroxylation에 의한 번역 후 변형을 통해 조절됩니다. HIF-1은 HIF-1 alpha와 HIF-1 beta의 heterodimer로 구성되며, 두 서브유닛 모두 지속적으로 번역됩니다.
정상 산소 조건(normoxia)에서는 HIF-1 alpha가 hydroxylation되어 polyubiquitination 후 proteasome에서 분해되지만, 저산소 조건(hypoxia)에서는 이 과정이 억제되어 HIF-1 alpha의 농도가 증가하고 전사 활성도가 높아집니다. 반면 HIF-1 beta는 산소 조건에 관계없이 안정적으로 존재합니다.
HIF-1 beta 기능 이상은 hypoxia 및 신세포암(renal cell carcinoma)과 관련이 있습니다.
