The epitope recognized by this antibody is reported to be sensitive to formaldehyde fixation and tissue processing.
We recommend the use of acetone fixation for frozen sections.
For FACS analysis, use 10 µL of the suggested working dilution to label 1×10^6 cells in 100 µL.
Rat anti-mouse CD8 alpha, clone KT15, recognizes the alpha chain of mouse CD8.
Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family that plays a key role in T cell signal transduction, differentiation, and activation.
CD8 is predominantly expressed on T cells as a disulfide-linked heterodimer of CD8α and CD8β, functioning as a co-receptor with T cell receptor (TCR) for MHC class I molecules.
CD8 is also present on subsets of T cells, NK cells, monocytes, and dendritic cells as CD8α homodimers.
Engagement of MHC-I/peptide complexes triggers recruitment of Lck kinase, leading to cytokine production, motility, and cytotoxic T lymphocyte (CTL) activation.
Activated CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
