Target Information

Vadastuximab is a biosimilar antibody targeting CD33, a transmembrane protein of the sialic acid-binding immunoglobulin-like lectin (Siglec) family.
CD33 contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit protein tyrosine phosphatases SHP-1 and SHP-2, and mediate receptor internalization via ubiquitin pathways.
It is expressed on cells of the myelomonocytic lineage and binds sialic acid residues in N- and O-glycans on cell surfaces. CD33 serves as a therapeutic and diagnostic marker for acute myeloid leukemia (AML) and is expressed on granulocyte and macrophage precursors in bone marrow, but not on pluripotent stem cells.
Additionally, CD33 is present on peripheral monocytes and is useful for distinguishing myelogenous leukemia cells from lymphoid or erythroid leukemias.
Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
