Target Information

Laprituximab is a biosimilar that targets EGFR.
EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1) is encoded by the EGFR gene located on chromosome 7 in humans.
EGFR belongs to the HER/ErbB family of receptor tyrosine kinases including ErbB2, ErbB3, and ErbB4.
Upon ligand binding (e.g., EGF, TGF-α), EGFR dimerizes and autophosphorylates tyrosine residues in the cytoplasmic domain, activating multiple downstream signaling cascades such as Ras–ERK, PI3K–Akt, Jak–STAT, and PKC pathways.
Phosphorylation at Y1086 enables GRB2 binding and MAPK pathway activation. EGFR activation leads to endocytosis and either degradation or recycling.
Mutations in EGFR are associated with lung cancer, and overexpression correlates with poor prognosis in various tumors including head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, and esophagus.
