Immunogen

This antibody was raised by immunising Wistar Furth rats with the mCCRL2 peptide (aa 1–24) conjugated to KLH.

Specificity:
This antibody is specific for murine CCRL2 (formerly known as L-CCR), expressed on mast cells, activated macrophages, and endothelial cells. CCRL2 binds the leukocyte chemoattractant chemerin without triggering classical GPCR signaling, enabling local enrichment of chemerin at inflammatory sites. CCRL2 presents active chemerin to leukocytes expressing the chemoattractant receptor CMKLR1/Chemr23. Chemerin can block anti-CCRL2 antibody binding.

Preparation:
The antibody was raised by immunising Wistar Furth rats with the mCCRL2 peptide/KLH conjugate. The immunizing amino-terminal mCCRL2 peptide sequence was NH2-MDNYTVAPDDEYDVLILDDYLDNSC-COOH, corresponding to residues 1–24 of mCCRL2, with a nonnative carboxyl-terminal cysteine to facilitate conjugation to keyhole limpet hemocyanin (KLH).

Also known as chemokine receptor X, human chemokine receptor (HCR), CRAM-A or CRAM-B, CCRL2 is a seven-pass transmembrane protein expressed by monocytes, neutrophils, and dendritic cells of immune tissues (primarily spleen, fetal liver, bone marrow, and lymph node). It shares over 40% homology with other C-C chemokine receptors but lacks the conserved DRY motif essential for signaling and ligand-binding. C-C chemokine receptors are G-protein coupled, seven-pass transmembrane proteins that function primarily in the chemotaxis of T cells and phagocytic cells to areas of inflammation.
