Product Specific Information

The 1C10 monoclonal antibody reacts with mouse CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a TNFR family member expressed by mouse B lymphocytes, follicular dendritic cells, thymic epithelium, and some peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype switching and preventing apoptosis, promoting proliferation.

Interaction between CD40 and CD154 (gp39) on T cells is critical for T-B cell crosstalk and immune regulation.
The 1C10 monoclonal antibody has agonistic activity in vitro and in vivo.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody titration recommended for optimal performance.

Light Sensitivity: Tandem dye sensitive to photo-induced oxidation. Protect from light.
Fixation: Samples may be fixed using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm; Emission: 667 nm; Laser: Blue, Green, Yellow-Green.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD40 is a single-chain glycoprotein of the TNF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells, and also on epithelial cells, carcinomas, and dendritic cells. CD40 mediates immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

AKNA transcription factor regulates CD40 expression, influencing cell interactions. TNFR2 adaptor protein mediates CD40 signal transduction. CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, an early event in Alzheimer’s disease pathogenesis. Two alternatively spliced transcript variants exist.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
