Product Specific Information

Description:
The HIT2 monoclonal antibody reacts with the human CD38 molecule, an approximately 45 kDa type II transmembrane protein. CD38 is expressed by thymocytes, peripheral B cells including plasma cells, activated T cells, and monocytes. CD38 serves as a counter-receptor for CD31 and exhibits cyclase and hydrolase enzymatic activity.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark. Clone-specific performance should be determined empirically.

Excitation / Emission / Laser:
488–561 nm excitation; 667 nm emission; compatible with Blue, Green, and Yellow-Green lasers.

Filtration:
0.2 µm post-manufacturing filtered.

Target Information

CD38 (NAD⁺ glycohydrolase) is a type II transmembrane glycoprotein that induces activation, proliferation, and differentiation of mature lymphocytes and mediates apoptosis of progenitor cells. It functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD⁺ glycohydrolase) and as a cell surface receptor.

Antibodies to CD38 are useful for:

• Subtyping lymphomas and leukemias
• Detecting plasma cells (e.g., myeloma identification)
• Marking activated B and T cells

CD38 also participates in cell adhesion, signal transduction, and calcium signaling. It is expressed at high levels in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma.
Diseases associated with CD38 dysfunction include chronic lymphocytic leukemia and Richter’s Syndrome.
