Product Specific Information

The HI100 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subsets of CD4+ and CD8+ peripheral T lymphocytes and B cells. CD45RA+ T cells are mainly naive/virgin, making HI100 useful as a phenotypic marker to discriminate T cell subsets.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Pre-titrated and validated for flow cytometry of normal human peripheral blood cells. Use at 5 µL (0.125 µg) per test (100 µL final volume).
• Light Sensitivity: Tandem dye is photo-sensitive; protect from light.
• Fixation: Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
• Excitation/Emission: 488–561 nm / 667 nm; compatible with Blue, Green, and Yellow-Green lasers.
• Filtration: 0.2 µm post-manufacturing filtered.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid cell subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells and regulates Src-family kinases, lymphocyte development, and antigen signaling. It exists in multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. CD45 is vital for signal transduction and immunological regulation. Dysfunction is associated with severe combined immunodeficiency and Hepatitis C.
