Antibody detects endogenous levels of CDC34 only when phosphorylated at Ser71.

CDC34 accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins.

Catalyzes Lys-48–linked polyubiquitination in vitro.
Cooperates with E2 UBCH5C and SCF (FBXW11) E3 ligase complex for polyubiquitination of NFKBIA, leading to proteasomal degradation.
Performs ubiquitin chain elongation from UBE2D3-primed NFKBIA-linked ubiquitin.
Regulates cell proliferation via SCF (SKP2) E3 ligase complex through ubiquitination and degradation of MYBL2 and KIP1.
Involved in degradation of CREM isoform ICERIIgamma and ATF15, abrogating their repression of cAMP-induced transcription.
Regulates G2/M phase by targeting WEE1 kinase for ubiquitination and degradation.
Also involved in degradation of beta-catenin.
Target of human herpes virus 1 protein ICP0, leading to ICP0-dependent interaction with proteasomes.
