Target Information
RIPK1은 세린/트레오닌 키나아제로, 세포사멸 수용체의 활성화, 병원체 인식 수용체(PRRs) 자극, DNA 손상 등에 의해 염증 및 세포사멸 신호를 전달합니다. TNF-α 수용체(TNFR1)가 활성화되면 TRADD와 TRAF2가 수용체에 결합하여 DAB2IP를 Ser-728에서 인산화하고, MAP3K5-JNK 세포사멸 경로를 활성화합니다. TRAF2에 의한 Lys-63 연결형 유비퀴틴화는 MAPK 및 NF-κB 경로의 하위 신호전달을 강화하며, 이는 염증 관련 유전자 발현을 유도합니다. RIPK1은 RIPK3와 결합하여 괴사(necrosis)를 유도하는 복합체를 형성하기도 합니다.
