Product Specific Information
This NJTEN3 monoclonal antibody reacts with the pro-form of mouse IL-1 beta, a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. It recognizes only the inactive 31 kDa pro-form and not the active (cleaved) form.
- Applications Reported: Suitable for intracellular staining and flow cytometric analysis.
- Applications Tested: Tested on stimulated mouse thioglycolate-elicited peritoneal macrophages using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
- Recommended protocol: BestProtocols® – Protocol A
- Use ≤ 0.125 µg per test in 100 µL final volume.
- Cell count: 10⁵–10⁸ cells/test (empirically determined).
- Antibody titration recommended for optimal performance.
Fluorochrome Information:
APC-eFluor® 780 emits at 780 nm and is excited with the Red laser (633 nm). Ensure instrument compatibility.
Light-sensitive: Protect vial and samples from light.
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark.
Excitation: 633–647 nm; Emission: 780 nm; Laser: Red.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
Interleukin-1 beta (IL-1β) is a proinflammatory cytokine synthesized in response to inflammatory stimuli as a 31 kDa inactive pro-form. Activation of inflammasomes leads to cleavage into the active 17 kDa form via caspase-1 processing. IL-1β is secreted through a non-classical pathway and signals via IL-1RI and IL-1RII, shared with IL-1α. It plays a key role in innate immune responses and acute-phase reactions. Elevated levels are associated with chronic inflammatory diseases, making IL-1β or IL-1RA potential therapeutic targets.
