Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, and binds glycosylated, fucosylated, sulfated sialylated glycoproteins such as CD34, GlyCAM-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

• Tested by flow cytometry on normal human peripheral blood cells
• Recommended use: 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Compatibility Notes

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI)
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays

CellBlox Plus Blocking Buffer

• Required for optimal staining with NovaFluor conjugates
• Add 5 µL per sample before combining with cells
• For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor of the selectin family. It includes:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer region
• Transmembrane region
• Short cytoplasmic region

CD62L is constitutively expressed on all classes of leukocytes including lymphocytes (except memory T cells), monocytes, and polymorphonuclear cells.
