Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, and binds glycosylated, fucosylated, sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.2 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL (10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability detection.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation for optimal staining.
Add 5 µL per sample (regardless of the number of NovaFluors) to antibody cocktails before combining with cells.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology, utilizing nucleic acid dye structures engineered for minimal spillover and spread impacts.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor of the selectin family. It consists of:

• Aminoterminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer region, transmembrane region, and cytoplasmic region

CD62L is constitutively expressed on leukocytes including lymphocytes (excluding many memory T cells), monocytes, and polymorphonuclear cells.
