Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells, binding glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1. These interactions mediate lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells at 5 µL (0.2 µg)/test.
A test is defined as the antibody amount staining a 100 µL cell sample (10⁵–10⁸ cells/test).

NovaFluor Compatibility:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Recommended: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

CellBlox Plus Blocking Buffer Usage:
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktails before combining with cells.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered fluorescent signatures minimizing spillover and spread impacts.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor of the selectin family. It includes:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on all leukocyte classes including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
