Product Specific Information

The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 acts as a ligand for CD80 (B7-1) and CD86 (B7-2), serving as a potent co-stimulator of T cell activation. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and increases cytotoxicity of CD3-activated T cells.

Each product includes:

  • 1 vial of CellBlox Plus Blocking Buffer

Applications Tested

  • Pre-titrated for flow cytometric analysis of normal human peripheral blood cells
  • Recommended use: 5 µL (0.8 µg) per test in 100 µL final volume
  • Cell number range: 10⁵–10⁸ cells/test

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence and minimal spectral spillover.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell surface glycoprotein, belonging to the immunoglobulin superfamily. It is constitutively expressed on most peripheral T lymphocytes and serves as a key co-stimulatory receptor by binding CD80 and CD86 on antigen-presenting cells. CD28 signaling prevents T cell anergy and apoptosis, supporting immune activation.

High expression is found in thymocytes and immature T cells, with lower levels in mature CD4⁺ and CD8⁺ T cells. CD28 dysfunction is associated with diseases such as mycosis fungoides and Sezary’s disease.
