Product Specific Information

This rabbit polyclonal antibody reacts with mouse and rat Receptor Interacting Protein (RIP) 3, a 57 kDa Ser/Thr kinase expressed ubiquitously in the cytoplasm. RIP3 can shuttle between nucleus and cytoplasm and binds to and phosphorylates RIP1, inhibiting RIP1- and TNFR1-mediated NFκB activation. It mediates programmed necrosis through regulation of TNF-induced reactive oxygen species production and acts as a molecular switch between TNF-induced apoptosis and necrosis.

Applications Reported: Western blotting and immunohistochemistry.
Applications Tested: Validated by western blot analysis using NIH-3T3 cell lysate (0.5–4 µg/mL recommended). Optimize titration for best assay performance.

Target Information

RIPK3 is a serine/threonine kinase of the receptor-interacting protein family, possessing a unique C-terminal domain. It is mainly localized in the cytoplasm and mitochondria, capable of nucleocytoplasmic shuttling via specific localization signals. RIPK3 is part of the TNF receptor-I signaling complex, inducing apoptosis and weak NF-κB activation. Caspase-8 prevents RIPK3-dependent necrosis by forming a proteolytically active complex with CFLAR, essential for protective function.
