Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that connect the TNF receptor superfamily to intracellular signaling pathways. They play crucial roles in regulating cell death and stress responses. TRAF proteins share a conserved region that mediates receptor binding and interaction with other TRAFs, enabling activation of various signaling cascades.
For instance, TRAF2 and TRAF3 bind directly to CD40, a receptor involved in B cell immunity, and are essential for NF-κB activation in mouse B lymphocytes. TRAF2 together with TRAF6 is required for CD40 signaling in non-hematopoietic cells. Additionally, TRAF2 interacts with the lymphotoxin-beta receptor (LTβR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
