Target Information
Estrogen Receptors (ER) belong to the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor functions as a ligand-activated transcription factor that, upon binding to estrogen, undergoes a conformational change enabling dimerization and binding to estrogen response elements (ERE) in DNA. This binding can lead to positive or negative regulation of gene transcription through recruitment of coactivator or corepressor proteins.
Two main forms of estrogen receptors exist — alpha (ESR1) and beta (ESR2) — encoded by separate genes. Alternative RNA splicing produces at least four known estrogen receptor-alpha isoforms:
- Isoform 1 (66 kDa)
- Isoform 2 (53 kDa)
- Isoform 3 (47 kDa)
- Isoform 4 (35 kDa)
Estrogen receptors are widely expressed in various tissues, essential for sexual development and reproductive function, and play roles in bone and other tissues. They are implicated in pathological processes such as breast cancer, endometrial cancer, and osteoporosis.
