• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

CD73 (Ecto-5-prime-nucleotidase, 5-prime-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5-prime mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
CD73 is a dimer of two identical 70 kDa subunits anchored to the plasma membrane via a glycosyl phosphatidyl inositol linkage. It serves as a marker of lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases.
Other forms of 5-prime nucleotidase exist in the cytoplasm and lysosomes and differ from CD73 by substrate affinities, magnesium ion requirements, ATP activation, and phosphate inhibition.
The CD73 gene is localized to chromosome 6q14–q21. Defects in this gene can lead to joint and arterial calcification, as well as intestinal tuberculosis. Two transcript variants encoding different isoforms of CD73 have been identified.
