Product Specific Information

The HM48-1 monoclonal antibody reacts with mouse CD48 antigen (also known as BCM1, Blast-1 in human, and OX-45 in rat). CD48 is a 45 kDa GPI-linked glycoprotein of the SLAM and Ig superfamily, expressed on most hematopoietic cells. It plays a critical role in adhesion and T cell activation.

Hematopoietic stem and progenitor cells show differential expression of SLAM family members (CD48, CD150, CD244), which helps distinguish their functional states:

• HSCs: CD150(+)CD244(-)CD48(-)
• MPPs: CD244(+)CD150(-)CD48(-)
• Restricted progenitors: CD48(+)CD244(+)CD150(-)

HM48-1 modulates CD48 function by blocking CD48/CD2 and CD48/CD244 interactions, inhibiting spleen cell proliferation, providing T cell costimulation, and prolonging cardiac allograft survival in vivo.

Applications Tested: Tested on mouse splenocytes at ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Target Information

CD48 (Blast-1) belongs to the CD2 subset of the Ig superfamily (CD2, CD58, CD84, CD150, CD229, CD244, etc.). It is a GPI-anchored protein broadly expressed on hematopoietic cells and serves as a high-affinity ligand for 2B4 and a low-affinity ligand for CD2.
2B4–CD48 interactions regulate NK and NK-T cell proliferation. CD48 signaling activates eosinophils and is elevated in infectious diseases. It is strongly expressed on lymphocytes and monocytes, weakly on granulocytes, and absent on platelets, fibroblasts, epithelium, and endothelium.
CD48 is a marker for detecting GPI-anchor defects (e.g., PNH) and is implicated in allergic airway diseases, making it a potential therapeutic target for allergic rhinitis and asthma.
