Product Specific Information

Description:
The HI30 monoclonal antibody reacts with all isoforms of human CD45, also known as Leukocyte Common Antigen (LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase enzymatic activity and plays an important role in activation of lymphocytes.

Applications Reported:
The HI30 antibody has been reported for use in flow cytometric analysis.

Applications Tested:
The HI30 antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. It can be used at ≤0.25 µg per test (defined as staining a 100 µL cell sample). Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase (PTP) expressed in all nucleated hematopoietic cells, comprising approximately 10% of all surface proteins in lymphocytes. It is absent on non-hematopoietic cell lines and tissues. CD45 glycoprotein regulates lymphocyte development and antigen signaling, acting as a key modulator of Src-family kinases.
CD45 protein exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. Isoforms show cell-type and physiological state-dependent expression and influence membrane domain localization.
CD45 regulates T- and B-cell receptor signaling, modulates JAK kinase activity, and impacts cytokine receptor signaling. It also contributes to integrin-mediated signal transduction, apoptosis regulation, and immune function modulation.
