Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress responses. TRAF proteins share a conserved homology region allowing receptor and TRAF–TRAF interactions, activating various signal cascades.
TRAF2 and TRAF3 bind directly to CD40, a NF receptor superfamily member crucial for NF‑κB activation in B lymphocytes. TRAF2 and TRAF6 are required for CD40 signaling in nonhemopoietic cells. TRAF2 also interacts with lymphotoxin‑beta receptor (LTβR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
