Target Information
Tumor necrosis factor (TNF) receptor associated factors (TRAFs) are adaptor proteins that link the TNF receptor superfamily to signaling pathways, regulating cell death and stress response. TRAF proteins share a homology region enabling receptor and TRAF–TRAF interactions, activating distinct signaling cascades.
TRAF2 and TRAF3 bind directly to CD40, a NF receptor superfamily member crucial for NF-kappa-B activation in mouse B lymphocytes. TRAF2 and TRAF6 are required for CD40 signaling in nonhemopoietic cells. TRAF2 also interacts with the lymphotoxin-beta receptor (LTbetaR) along with TRAF3 and apoptosis inhibitors cIAP1 and Smac.
