Product Specific Information

The monoclonal antibody reacts with human AITR (Activation Inducible TNFR family member), an approximately 25 kDa member of the TNFR superfamily.
AITR mRNA is detected in lymph node, peripheral blood leukocytes, and weakly in spleen. Expression increases within 24 hours after PMA/ionomycin or PHA stimulation of PBMC.
At the protein level, AITR is expressed by a small population of activated PBMC. It associates with TRAF1, TRAF2, and TRAF3 and induces NF-kappaB activation via TRAF2.
The ligand TL6 (AITRL) interacts with AITR and mediates cross-talk between T lymphocytes and endothelial cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested on PHA-stimulated normal PBMC. Use at 5 µL (0.25 µg) per test (100 µL final volume). Cell number typically ranges from 10⁵ to 10⁸ cells/test.
Excitation: 488 nm; Emission: 519 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

Tumor necrosis factor receptor superfamily member 18 (TNFRSF18), also known as GITR or AITR, is encoded by the TNFRSF18 gene (chromosome 1p36.33).
It is a TNF-receptor family protein whose expression increases upon T-cell activation and plays a key role in immunological self-tolerance maintained by CD25⁺CD4⁺ regulatory T cells.
Knockout studies in mice suggest involvement in CD3-driven T-cell activation and programmed cell death.
Three alternatively spliced transcript variants encoding distinct isoforms have been reported.
