Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.1-expressing mouse strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed by all leukocytes in these strains.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes at ≤1.0 µg/test (100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It plays a crucial role in lymphocyte development and antigen signaling by regulating Src-family kinases.
CD45 exists as multiple isoforms via alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains. Isoform expression varies by cell type and physiological state.
CD45 also influences cell survival through integrin-mediated signaling and participates in DNA fragmentation during apoptosis. CD45RA is one such isoform with restricted expression among lymphoid subtypes.
