The homeobox DNA-binding domain is a 60-amino-acid motif conserved among species, functioning via a helix-turn-helix structure to regulate transcription and gene expression.
PRRX1 (paired related homeobox 1), also known as PMX1 or PHOX1, is a 245-amino-acid protein containing one OAR domain and one homeobox DNA-binding domain, belonging to the paired homeobox family.
Localized in the nucleus, PRRX1 acts as a transcriptional co-activator enhancing DNA-binding activity of serum response factor (SRF), mediating SRF-dependent gene expression induced by growth and differentiation factors.
Additionally, PRRX1 regulates transcriptional activity of creatine kinase-M (muscle), contributing to mesodermal muscle type establishment.
PRRX1 exists as two alternatively spliced isoforms: PMX1-A and PMX1-B.
