Target Information
TRIM5 is a member of a broad family of proteins characterized by a tripartite motif containing a RING domain, B-box type 1, and B-box type 2, followed by a coiled-coil region. TRIM5 has five alternately spliced isoforms; the alpha variant includes a carboxy-terminal B30.2 (SPRY) domain. Although the function of TRIM5-gamma is not fully defined, TRIM5-alpha variants from humans and other primates confer resistance to various retroviruses, including HIV-1, with varying efficiencies. The TRIM5δ isoform serves as a scaffold for BTBD1 and BTBD2 assembly and exhibits autoubiquitination activity dependent on RING finger and UbcH5B.
