Product Specific Information

The monoclonal antibody BB27 reacts with human CD101 (V7 antigen), a member of the Ig superfamily. CD101 is a disulfide-linked homodimer expressed on monocytes, granulocytes, and dendritic cells (Langerhans-like cells expressing HLA-DR, CD1a, CD1c). It is also expressed on T lymphocytes, where it plays a role in cell activation. CD101⁺CD28⁺ cells are highly responsive to CD28 signaling. In combination with anti-CD28 or suboptimal levels of anti-CD3, anti-CD101 can enhance proliferation, suggesting an activating role.

The BB27 antibody has been shown to inhibit T-cell reactivity in allogeneic and antigen-specific mixed DC-T cell cultures. In mice, CD101 is expressed on a subpopulation of regulatory T cells (CD4⁺, CD25⁺, Foxp3⁺) with high suppressor activity. In human PBMCs, CD101 staining is observed in approximately 30% of Foxp3⁺ cells, though enhanced suppressor activity has not been confirmed in humans.

A test is defined as the amount of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal assay performance.

Target Information

CD101 (V7 antigen) is a disulfide-linked homodimer of unknown function, expressed on monocytes, granulocytes, dendritic cells, and T lymphocytes. Its expression on T cells is associated with activation responses. CD101⁺CD28⁺ T cells show enhanced CD28-mediated signaling, and anti-CD101 can augment proliferation when combined with anti-CD28 or suboptimal anti-CD3. In mice, CD101 marks a subset of regulatory T cells with strong suppressor activity, while in humans, it is expressed on about 30% of Foxp3⁺ PBMCs without confirmed enhanced suppressor function.
