Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein. CD40 is a member of the TNFR family and is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells.
CD40 regulates B cell development and maturation by inducing Ig isotype-switching and, together with IL-4, protects B cells from apoptosis and promotes proliferation. Interaction of CD40 with CD154 (gp39), its ligand on T cells, is critical for T-B cell crosstalk and immune regulation.

Target Information

CD40 is a single-chain glycoprotein, part of the TNF receptor superfamily, homologous to CD30. It is expressed on all B cells except plasma cells and also found on epithelial cells, carcinomas, and lymphoid dendritic cells.
CD40 is essential for immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.
AKNA transcription factor and TNFR2 adaptor protein are known to regulate CD40-mediated signaling. CD40-CD154 interaction is implicated in amyloid-beta-induced microglial activation, suggesting a role in Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
