• Store vial at -20°C prior to opening.
• Aliquot contents and freeze at -20°C or below for long-term storage.
• Avoid repeated freeze/thaw cycles.
• Centrifuge product if not completely clear after standing at room temperature.
• Stable for several weeks at 4°C as an undiluted liquid.
• Dilute only prior to immediate use.

The antiserum is directed against mature 17,000 MW mouse TNFα and is useful in determining its presence in various assays.
The antibody does not recognize mouse TNFβ (lymphotoxin).
It recognizes the cell-bound precursor of TNFα (26,000 MW protein) in immunoblots, particularly in denatured samples.
This antibody is also useful for neutralization of mouse TNFα activity in bioassays but does not neutralize lymphotoxin activity.
For neutralization, incubate the sample with a 1:200 dilution of the antibody for at least 4 hours before testing.
A control of similarly diluted normal rabbit IgG is recommended.

Target Information

TNF alpha is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF alpha regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It exists as multimers (dimers, trimers, or pentamers) but appears as a single 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice suggest neuroprotective functions and tumor necrosis induction in tumor-bearing mice.
TNF alpha is involved in immune responses to bacterial, viral, parasitic, and fungal infections, and plays a role in tumor necrosis.
It acts synergistically with interferon-gamma in cytotoxicity and is implicated in autoimmune diseases, insulin resistance, and cancer.
