Target Information
Members of the Id family of basic helix-loop-helix (bHLH) proteins include ID1, ID2, ID3, and ID4. These proteins are ubiquitously expressed and form dimers with class A and B HLH proteins. Due to the absence of the basic region, ID proteins cannot bind DNA and thus act as negative regulators of DNA binding by bHLH proteins. ID1 inhibits DNA binding of E12 and Myo D, thereby suppressing muscle-specific gene expression. During muscle cell differentiation, ID protein levels decrease, allowing E12/E47 to interact with Myo D and myogenin, activating myogenic differentiation. Expression of ID proteins is growth factor-dependent, and reduction of ID mRNA delays cell cycle reentry after growth factor stimulation.
