Target Information

TLR4 is a member of the Toll-like receptor (TLR) family, playing a fundamental role in pathogen recognition and activation of innate immunity. TLRs are highly conserved from Drosophila to humans and share structural and functional similarities.
This receptor is most abundantly expressed in placenta and myelomonocytic leukocyte subpopulations. Mammalian cells respond to LPS by activating TLR4, which is part of a multi-protein complex including CD14, LY96, and TLR4.
TLR4 mediates signal transduction induced by lipopolysaccharide (LPS) found in gram-negative bacteria and aids in recognition of pathogen-associated molecular patterns (PAMPs). Mutations in TLR4 gene are associated with differences in LPS responsiveness.
TLR4 is expressed by peripheral blood monocytes, a small population of B-cells, and human placenta. TLR4-deficient mice show increased susceptibility to Gram-negative sepsis.
