Target Information
Prolyl hydroxylase 4 (P4HTM) is a prolyl hydroxylase that modifies HIF-alpha. Classic prolyl hydroxylases are found in the endoplasmic reticulum and modify collagen, whereas HIF is an intracellular protein with distinct hydroxylation sites. HIF acts as a transcriptional complex critical for oxygen homeostasis. Prolyl hydroxylase 4 catalyzes the formation of 4-hydroxyproline in HIF-alpha proteins, functioning as a cellular oxygen sensor. Under normoxic conditions, it hydroxylates HIF-alpha, targeting it for proteasomal degradation via the von Hippel-Lindau ubiquitylation complex.
