Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3 is a 49–55 kDa transcriptional regulator identified as the gene defective in scurfy (sf) mice. It is highly expressed in CD4⁺CD25⁺ regulatory T cells (Treg), and ectopic expression in CD4⁺CD25⁻ cells induces a Treg phenotype.

Epitope mapping shows recognition of amino acids 75–125 of mouse Foxp3. This region is alternatively spliced in humans. FJK-16s is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Use of other buffers is not recommended.

Applications Tested:
Tested in intracellular flow cytometry of mouse splenocytes using Anti-Mouse Foxp3 FITC Staining Set (Product #71-5775-40).
Recommended ≤1 µg/test (100 µL final volume). Cell count: 10⁵–10⁸ cells/test.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue.
Filtered through 0.2 µm post-manufacturing.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is expressed in CD25⁺CD4⁺ regulatory T cells and absent in CD8⁺ T cells. FOXP3 defects cause the scurfy phenotype in mice and IPEX syndrome in humans. Transcript variants encoding different isoforms exist. In human breast and colon cancer cells, FOXP3 expression is regulated by p53 in response to DNA damage.
