Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. The receptor is larger than 90 kDa and has three major functional domains:

  • N-terminal domain
  • DNA-binding domain
  • Androgen-binding domain

AR acts as a ligand-activated transcription factor that binds active testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, the receptor dissociates from accessory proteins, translocates into the nucleus, dimerizes, and stimulates transcription of androgen-responsive genes.

The AR signaling pathway is essential for development and function of male reproductive organs (prostate, epididymis) and also plays roles in nonreproductive tissues such as muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase signaling; inhibition of MEK1/2 correlates with changes in AR phosphorylation state.

Abnormalities in AR signaling are associated with diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations can lead to complete androgen insensitivity (CAIS).
