Product Specific Information

The BM8 monoclonal antibody reacts with mouse F4/80 antigen, an approximately 160 kDa surface receptor belonging to the EGF-TM7 family. It contains seven EGF-like domains, seven transmembrane regions, and an intracellular domain homologous to other TM7 proteins. F4/80 is highly expressed on mature macrophages, serving as a key marker for this population. Other cell types such as eosinophils, Langerhans cells, and certain dendritic cells may also express F4/80.

Expression begins early in myeloid development and can be upregulated by M-CSF stimulation. Some macrophage subsets, particularly in lymphoid tissues, may lack F4/80 expression.

Applications Reported: Suitable for flow cytometry, IHC (frozen and paraffin sections), and WB under non-reducing conditions. Proteinase treatment is required for paraffin sections. Reduction with 2-mercaptoethanol destroys the BM8 antigen.

Applications Tested:

• Flow cytometry on mouse spleen or bone marrow cells (≤0.5 µg/test).
• IHC on formalin-fixed paraffin-embedded mouse tissue (≤10 µg/mL, pronase treatment recommended).
  Antibody titration is advised for optimal results.

Target Information

F4/80 antigen is a 160 kDa cell surface glycoprotein in the EGF-TM7 family, sharing 68% amino acid identity with human EMR1. Expression varies with macrophage maturation and activation. It is present on macrophages in various tissues (Kupffer cells, microglia, peritoneal, lung, thymus, bone marrow stroma, and spleen red pulp) and on some dendritic cell subsets. Absent in T-cell zones of spleen and lymph nodes. Recent studies suggest F4/80 may play a role in generating efferent CD8⁺ regulatory T cells.
