Product Specific Information
Description:
The AER-37 monoclonal antibody reacts with the Fc epsilon RI alpha subunit, an IgE-binding subunit lacking signal-transducing ability. FcεRIα is expressed on mast and basophil cells and is upregulated by the presence of IgE. It forms a tetrameric complex with one β and two γ subunits, which possess immunoreceptor tyrosine-based activation motifs (ITAM). The FcεRI complex plays an important role in triggering IgE-mediated allergic reactions.
Applications Reported:
This antibody has been reported for use in flow cytometric analysis.
Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.25 µg) per test in a final volume of 100 µL.
Cell number can range from 10⁵ to 10⁸ cells/test.
Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm).
Recommended filter: 710/50 bandpass (695/40 acceptable alternative).
Ensure instrument compatibility.
Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.

Target Information
FceR1 (Fc epsilonR1 alpha):
A subunit of the high-affinity receptor of IgE, forming a tetrameric complex with one α, one β, and two γ subunits. The β and γ subunits mediate signal transduction. This receptor is key in triggering allergic responses such as hay fever and asthma. Crosslinking of IgE-bound FcεRI receptors by allergens leads to release of histamine and other mediators, initiating inflammatory responses.
