Target Information

XBP1 encodes a transcription factor that regulates MHC class II genes by binding to a promoter element referred to as an X box. This gene product is a bZIP protein, which also binds to an enhancer in the promoter of the T cell leukemia virus type 1 promoter, potentially increasing viral protein expression by acting as a DNA-binding partner of a viral transactivator.

When unfolded proteins accumulate in the endoplasmic reticulum (ER), the mRNA of XBP1 is processed into an active form by an unconventional splicing mechanism mediated by the endonuclease IRE1. This splicing removes 26 nucleotides, resulting in a frame-shift and the formation of isoform XBP1(S), the functionally active transcription factor. The unspliced isoform, XBP1(U), acts as a negative feedback regulator of XBP1(S), shutting off transcription of target genes during ER stress recovery. A pseudogene of XBP1 has been identified on chromosome 5.
