Target Information
PAI1 (plasminogen activator inhibitor 1)은 세린 프로테아제 억제제(superfamily of serine protease inhibitors)에 속하며, 조직형 및 유로키나제형 플라스미노겐 활성화제(tPA, uPA)의 주요 억제제입니다.
혈소판이 순환 중 PAI1의 주요 공급원이지만, 섬유아세포, 평활근세포, 내피세포, 간세포, 염증세포 등 다양한 조직과 세포에서 합성 및 분비됩니다.
PAI1 발현은 성장인자, 사이토카인, 호르몬, 염증 인자, 대사 산물, 혈관 조절 인자, 화학물질, 물리적 요인 등 다양한 요소에 의해 전사 수준에서 조절됩니다.
비만, 혈전증, 대사증후군 등 여러 질환 상태에서 PAI1 수치가 증가하며, 혈전 발생과 연관되어 있습니다.
PAI1 결함은 이상 출혈을 특징으로 하며, 플라스미노겐 활성화제의 활성을 억제하여 섬유소 용해(fibrinolysis)를 억제하고 신경 세포 생존에 기여할 수 있습니다.
또한, PAI1 결핍(plasminogen activator inhibitor-1 deficiency)의 원인이 되며, 다양한 전사 변이체가 보고되어 있습니다.
