Product Specific Information
Despite the predicted molecular weight, Siglec11 often migrates at a higher molecular weight in SDS-PAGE, presumably due to post-translational modifications.
A suggested positive control is HepG2 cell lysate.
PA5-20743 can be used with blocking peptide PEP-0857.
Target Information
Siglecs are sialic acid-binding lectins of the immunoglobulin superfamily mainly expressed in cells of the hematopoietic system.
Siglec11 is unique in that it binds specifically to alpha2-8-linked sialic acids and is expressed on macrophages of various tissues rather than peripheral blood leukocytes.
It is highly homologous to Siglec10 over their extracellular domain but not over their cytoplasmic domain, suggesting Siglec11 arose through gene duplication and recombination involving another ancestral siglec gene.
Following pervanadate treatment, Siglec11 becomes tyrosine-phosphorylated and associates with SHP-1 and SHP-2.
At least five isoforms of Siglec11 are known to exist.
