Specificity Comments:
Recognizes a protein of 30–33 kDa identified as CD20. Expression is restricted to normal and neoplastic B-cells, absent from other leukocytes and tissues. CD20 is expressed by pre-B cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells.
Useful for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood, and B-cell localization in tissues. Reacts with most B-cells in peripheral blood and lymphoid tissues and their derived lymphomas. Germinal center blasts and B-immunoblasts are particularly reactive. Rare CD20-positive T-cell lymphomas and reactivity with Reed-Sternberg cells in Hodgkin’s disease have been reported.
Expected to react with Monkey, Baboon, Gorilla, and Chimpanzee.

Target Information

CD20 is a non-glycosylated surface phosphoprotein (33–37 kDa) expressed on mature and most malignant B cells, a subpopulation of T lymphocytes, and follicular dendritic cells. It regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, with intensity dependent on extracellular triggering and BCR aggregation. After receptor ligation, BCR and CD20 colocalize and then rapidly dissociate before BCR endocytosis, while CD20 remains at the cell surface.
CD20 is a key therapeutic target for antibody-mediated B-cell depletion in malignancies, as it remains membrane-bound after treatment.
Diseases associated with CD20 dysfunction include MS4A1-related common variable immune deficiency.
