Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation. It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells.
CD20 expression synchronizes with surface IgM and regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
It associates with lipid rafts, with intensity depending on extracellular triggers and BCR aggregation. Following receptor ligation, BCR and CD20 colocalize and then dissociate before BCR endocytosis, while CD20 remains on the cell surface.
CD20 is a key target for antibody-mediated B-cell depletion therapy, as it is highly expressed on B-cell malignancies and remains on the plasma membrane after treatment.
Diseases associated with CD20 dysfunction include MS4A1-related common variable immune deficiency.
