Target Information
CD20 is a non-glycosylated surface phosphoprotein with a molecular weight of 33–37 kDa depending on its phosphorylation state. It is expressed on mature and most malignant B cells, as well as on a subset of T lymphocytes and follicular dendritic cells. CD20 expression coincides with surface IgM and plays a role in calcium conductance, cell cycle progression, and B-cell proliferation.
CD20 is associated with lipid rafts, and its interaction with BCR (B-cell antigen receptor) changes dynamically upon receptor ligation. It remains on the cell surface after BCR endocytosis, making it a valuable target for antibody-mediated B-cell depletion therapies. High expression on B-cell malignancies and stability on the plasma membrane contribute to its therapeutic relevance.
Diseases associated with CD20 dysfunction include MS4A1-related common variable immune deficiency.
