• Immunohistochemistry (PFA fixed):
  Incubate antibody for 30 min at room temperature.
  Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

CD20 is a non-glycosylated surface phosphoprotein with a molecular weight range of 33–37 kDa depending on phosphorylation.
It is expressed on mature and most malignant B cells, a subset of T lymphocytes, and follicular dendritic cells.
CD20 regulates transmembrane calcium conductance, cell cycle progression, and B-cell proliferation.
It is associated with lipid rafts, and its interaction with the B-cell antigen receptor (BCR) changes upon extracellular stimulation.
CD20 remains at the cell surface after BCR endocytosis and serves as a target for antibody-mediated B-cell depletion.
High expression levels on B-cell malignancies make it useful for therapeutic applications.
Diseases associated with CD20 dysfunction include Ms4a1-related common variable immune deficiency.
