Target Information
PAI1 (plasminogen activator inhibitor 1)은 serine protease inhibitor superfamily에 속하며, tissue-type 및 urokinase-type plasminogen activators (tPA 및 uPA)의 주요 억제제입니다.
혈소판이 순환 중 PAI1의 주요 공급원이지만, 섬유아세포, 평활근세포, 내피세포, 간세포 및 염증세포 등 다양한 세포에서 합성·분비됩니다.
PAI1 발현은 성장인자, 사이토카인, 호르몬, 염증 인자, 대사물질, 혈관조절 인자 등 다양한 요인에 의해 전사 수준에서 조절됩니다.
여러 질병 상태에서 PAI1의 수준이 증가하며, 비만, 혈전성 질환 및 대사증후군과 관련된 혈전 발생 증가와 연관되어 있습니다.
PAI1 결함은 비정상적 출혈을 특징으로 하며, fibrinolysis 억제를 매개하여 plasminogen activator 활성을 억제하고 신경세포 생존을 촉진할 수 있습니다.
또한, 대체 스플라이싱된 전사 변이체들이 존재하여 다양한 isoform을 생성합니다.
